I've thought it would be nice to see it on the game map, too. Perhaps it does this but I've missed it, but for example if everyone is in the same area and someone marks the brush off to the side, it'll ping on the minimap but I don't know what exactly they're pinging unless it's a champion. Perhaps if it's on the ground a small semi-transparant flag could be place, with concentric rings radiating from it?

2. Again, I agree, for the most part, but it's a loading screen. I don't think it has to be totally awesome. I do wish it would be shorter though... When the loading screen comes up, I'm ready in about 3 seconds. I generally have to wait 30 seconds for others to be "ready". After everyone is ready, there's generally another 30 seconds wait before the game actually starts. I know a minute isn't a huge deal, but it feels eternal when you just watch bars not move. Maybe do a Pacman-esque loading screen. Have Rammus chasing Annie across the screen one way, then have him come the other way in ball form being chased by Tibbers and Annie.

LOL I like that idea! A bit overkill, but amusing. I'd settle for the ability to chat with teammates while the others that are still loading, load.

4. I have troubles linking heros with users. Someone says "Help me" and you have no idea which way to go because you don't know who is requesting. Chat needs to be " 2drk4u [Ryze] : Help me "

A great suggestion, too. Seems like people often refer to the champions rather than the users. E.g. "Annie, go bot" rather than "toonslayer go bot". Your suggestion would help put the two together.
Somewhat related, I'd like to see the two teams have different global chat colors. Often I see someone chat and wonder "is that guy on my team or the other?" since I almost exclusively play random games. I confess I have to check the scoreboard to verify. So anyway, make your own team's global chat blue or even a different color of green.
