I just used that and i dont know why and how, just puting them into combiner..... They are all somehow missing? Now are they activated auto or i cant use them anymore. I spend like lot of ip for that... Someone expain to me how that work and what did i do with that combiner?

It consumes (gets rid of) the runes you put into it, and gives you a random new rune. Look here:
